چکیده

Thymidylate synthetase activity of a human lymphoblastic cell line, CCRF-CEM, was elevated by the addition of methotrexate to the culture medium and was lowered when cytosine arabinoside was added. The simultaneous addition of both drugs resulted in levels of enzyme activity intermediate between those observed for each drug alone. The level of enzyme activity could be modulated by varying the ratio of the two drugs. The ratio of enzyme activity in homogenates of cells from control or drug-treated cultures remained constant with dialysis. Changes in the rate of thymidylate synthetase turnover are suggested as the basis for the variation of enzyme activity in the drug-treated cultures.

Depression of thymidylate synthetase activity in response to cytosine arabinoside.

Depression of thymidylate synthetase activity by cytosine arabinoside in CCRF-CEM cells was correlated with a blocking of precursor incorporation into RNA and with cell lysis. With increasing concentrations of cytosine arabinoside in the culture media, a proportional decrease of uridine incorporation into RNA and decrease in thymidylate synthetase activity occurred and was accompanied by lysis ...

Cytosine arabinoside and Methotrexate alter in different ways the metabolic patterns of leukocytes from patients with acute granulocytic leukemia. The variations in the incorpora tion into DNA of deoxyuridine and thymidine during the first twenty-four hours after cytosine arabinoside administration

Cytosine arabinoside and Methotrexate alter in different ways the metabolic patterns of leukocytes from patients with acute granulocytic leukemia. The variations in the incorpora tion into DNA of deoxyuridine and thymidine during the first twenty-four hours after cytosine arabinoside administration were correlated with one another but not with the variation in activity of thymidylate synthetase...

Impact of polyglutamation on sensitivity to raltitrexed and methotrexate in relation to drug-induced inhibition of de novo thymidylate and purine biosynthesis in CCRF-CEM cell lines.

The aim of this study was to investigate the influence of folylpolyglutamyl synthetase (FPGS) activity on the cellular pharmacology of the classical antifolates raltitrexed and methotrexate (MTX) using two human leukemia cell lines, CCRF-CEM and CCRF-CEM:RC2Tomudex. Cell growth inhibition and drug-induced inhibition of de novo thymidylate and purine biosynthesis were used as measures of the cel...

Decreased folylpolyglutamate synthetase activity as a mechanism of methotrexate resistance in CCRF-CEM human leukemia sublines.

Determinants of methotrexate (MTX) resistance in cell lines resistant to short, but not continuous, MTX exposure were investigated since such lines may have relevance to clinical resistance. CCRF-CEM R30dm (R30dm), cloned from CCRF-CEM R30/6 (a MTX-resistant subline of the CCRF-CEM human leukemia cell line), had growth characteristics similar to CCRF-CEM. R30dm was resistant to a 24-h exposure ...
